Temodar
Prodrug that spontaneously converts to MTIC at physiologic pH. Methylates DNA at O6-guanine and N7-guanine, causing cytotoxic DNA damage. Crosses blood-brain barrier.
Good CNS penetration — used for brain tumors. Myelosuppression (thrombocytopenia, neutropenia). Take on empty stomach. Monitor CBC. Used investigationally in dogs for glioma.
